چکیده
Many methods have been devised for producing gradients in elution fluids for ion-exchange chromatography (l-9). While many of these are invaluable for special purposes, none appears to allow: (a) the production of a complex multicomponent gradient directly from a graphic presentation, and (b) the possibility of changing from gradient to stepwise elution with the same system. A simple method for producing complex gradients or stepwise elutions is described.

Microchip emitter for solid-phase extraction-gradient elution-mass spectrometry.
A microchip electrospray emitter with a magnetic bead trap has been designed for solid-phase extraction-gradient elution-mass spectrometry (SPE-GEMS). The goal of this method is the detection of analytes at low concentrations and it is here demonstrated using reverse phase coated magnetic beads (Mbs) for the preconcentration and detection of the peptides. The sample is passed through the chip, ...
متن کاملGradient elution in microchannel electrochromatography.
There is great interest in using microfluidic channels packed with a stationary phase for chemical separations of complex mixtures. A key advantage of such techniques is the use of electroosmotic flow (EOF), controlled simply by applying electrical potentials between reservoirs. A disadvantage for this technique, however, is a lack of compatibility with gradient elution separations. This limita...
متن کاملHPLC: Isocratic or Gradient Elution and Assessment of Linearity In Analytical Methods
The aim of pharmaceutical analysis is to obtain the necessary qualitative and quantitative information about the investigated sample. HPLC is the dominant separation technique in modern pharmaceutical and biomedical analysis because it results in highly efficient separations and in most cases provides high detection sensitivity. Chromatographic optimization procedures are becoming more multidis...
متن کاملIsocratic and gradient elution chromatography: a comparison in terms of speed, retention reproducibility and quantitation.
Chromatographers are cautioned to avoid gradient elution when isocratic elution will do. In this work, we compared the analytical properties of gradient and isocratic separations of a sample which can be done quite readily under isocratic conditions. We found that gradient elution gave a shorter overall analysis with similar resolution of the critical pair compared to isocratic elution without ...
متن کاملSeparation and identification of urinary constituents by use of multiple-analytical techniques.
High-pressure anion-exchange chromatography with gradient elution has been used to resolve ultraviolet-absorbing constituents from normal and pathologic specimens of human urine. We have identified 50 of these constituents by using multiple-analytical techniques. Ultraviolet spectral maxima, gas chromatographic methylene unit (MU) retention values (for trimethylsilyl derivatives), and the major...
